The hosts extended their lead through Daniel Kasende to go into the break 21-10 up.
Finishing a flowing move for the hosts, Jack Walsh bagged Ospreys' bonus point in Swansea, before Kieran Hardy added another.
Max Pepper hit back for Newcastle Falcons but it wasn't enough to stage a comeback as Ospreys ran out 35-15 winners.
